The
Illumination
mode changes the laser power
(Fig. 64) to acquire a series of varying exposures.
The degree of variation is fixed to allow better
reproducibility of experiments. The second image is
acquired with 3 times more laser power and the
third image is acquired with 10 x more laser
power.
For the acquisition step with increased laser power,
those regions of the image are excluded from
exposures where no reasonable data can be
obtained with any higher laser power. This mask
reduces unwanted bleaching of the sample. The
respective ROIs are displayed in an additional
image (Fig. 65).
The calculation of the resulting image follows the
same procedures as for the sampling mode.
However, the regions, which are excluded from a
second and/or third illumination step are divided by
2 instead of 3.
