IMPORTANT NOTES FOR CHAPTER 4
Tile scan with
overlapping borders of
single images
When performing a tile scan with a defined overlap of > 0 the functions
Position
and
Stage
in the
2D View
tab /
Dimensions
are not available.
Changes to the
operating system
Do not update Windows Vista, the version installed from the factory is
already optimized to run with ZEN 2010. Automatic installation of patches
might lead to conflicts with ZEN.
Dye-spectra of
deselected channels
stay in the display
The spectra will stay until a new dye is chosen for this channel.
FCS model for PCH
In the PCH model decimals can be entered into the F1, F2 and F3 input boxes
the following way: for example instead of typing 0.361, type in 361, then
position the cursor before the digit 3, then enter the dot.
LSM 710 and LSM 780
with 32-channel
spectral detector
The light path will remain in lambda mode when the
Extract
function in the
unmixing image viewer is activated and one of the selected bands exceeds
the detection range of the LSM 710 / LSM 780 Quasar detector.
While the 'In Tune' laser is busy due to a wavelength change the wavelength
display is not marked in red as is indicated for NLO lasers (64-bit systems
only).
The automatic selection of the main dichroics in basic mode of the
Light
path
tool window (
Show all
mode not activated) can give an unexpected
result for the case of a line wise multi-track acquisition, where the 405 nm
laser, a line from the 'In Tune' laser and a line from one of the visible lasers
are selected. The main dichroics can block the light of the visible lasers.
In this case switch to
Show all
mode and select a neutral density beam
splitter. To achieve good laser suppression it is recommended to use the
frame-wise multi track mode.
LSM 710 and LSM 780
with 'In Tune' laser
The automatic selection of the main dichroics will consider the currently
selected wavelength of the 'In Tune' laser even if the laser is currently not in
use. This might block the laser wavelengths of other visible light laser lines.
The work-around is to tune the 'In Tune' laser to the lowest possible
wavelength.
Scan speed uni versus
bidirectional (for all
systems)
For some scan speeds the acquisition with two tracks in line wise multi-track
mode is faster in uni-directional mode than in bi-directional mode.
Switching and adding
of tracks
The switching between tracks and the adding of a new track in the
Illumination Setup
tool window in the left screen area works in a slower
mode compared to ZEN 2008.
Auto Save function
When a time series with the
Auto Save
function active and LSM-file format
selected is stopped manually during acquisition, the reloaded image series
will display arbitrary data instead of empty images for the frames which have
not been acquired.
