Position
In
Current Position
you can select a well from a carrier where you can perform a measurement. You can
also control the stage. When using this method, FCS measurements are performed on a fixed position
with parked scanners and without automatic table movement. In this case you can measure in solution or
by moving the structure of interest to the position manually under LSM control prior to FCS
measurements.
•
Click on the
Current Position
tab to open the
Current Position
panel (see Fig. 44). Click on any
other tab to leave the panel.
Selecting a well
•
Orient a carrier in the
Carrier Position
tool.
This will display the carrier (see Fig. 45). The
current selected well will be highlighted in blue.
•
Focus into the sample using the line scanning.
•
Select the carrier. The stage will move to the
selected well.
•
Perform Measurement by pressing the
Start
ConfoCor Experiment
action button.
The laser beam will stay at the optical axis,
because positioning of the well is done by
the scanning stage. If you want to have the
scanner position within the well, you have to
go to the LSM image panel, image the
solution and position by stage or scanning
mirrors.
Cross hair function
When using this method, FCS measurements are
performed on a fixed position without scanner or
stage. In this case the structure of interest must be
moved to the position manually under LSM control
prior to FCS measurements. If no offset or rotation
was defined the laser position with the parked
mirrors is the centre of the image. In other cases
the position of the laser beam with this rotation
within the image has to be determined by a bleach
experiment. The Crosshair is intended as an overlay
tool mark the position of the laser beam.
You cannot select a measurement position with the
Cross hair
; this position will not be
approached by the stage or the scanner. Instead you have to move your sample to the position of
the fixed laser beam (no stage or scanner movement).
