Motifs
Like in photography, common imaging conditions
with typical requirements also exist in laser
scanning microscopy.
The
Motifs
in
Smart Setup
represent presets of
scanning parameters (pixels size, pinhole diameter,
scan speeds etc.) for such typical imaging
conditions (Fig. 73):

Current:
Does not change the current set of scanning parameters defined in the Acquisition Tool.In the
Channels tool laser power and pinhole are set to moderate levels. Detector settings are not changed
unless the detector gain is still zero, then a moderate gain will be set. Clicking Apply does set the
parameters accordingly, no image is scanned yet.
Quality:
Aims to provide high quality images with best signal to noise ratio based on lower detector
gains and higher laser power defined in the Channels Tool. The pinhole is set to one Airy unit. In the
Acquisition tool the image size shows close to niquist settings in xy, a slow scan speed, 12 bit data depth
and unidirectional scan mode without averaging. Clicking apply sets the parameters and scans one
image.
Speed
: Choses parameters for fast imaging. The parameters of the Imaging Setup and the Channels tool
are not changed. Within the Acquisition tool the image size is reduced and using the maximum speed
possible for the given zoom with bidirectional scanning and no averaging an image is scanned when
clicking Apply.
Standard:
Applies the most common imaging settings for the acquisition.
The image size is set to 512 x
512 pixels with the standard scan speed of 9 at no average, unidirectional scanning and a data depth of
8 bit. Laser power is set to moderate levels and the gain is at rather low settings. Clicking apply sets the
parameters and scans a new image.
Widefield-Like:
Uses 512x512 pixels, no average, 12 bit data depth and unidirectional scanning at the
given imaging speed. The pinhole is fully opened and the gain and laser power are both set to low levels.
Clicking Apply sets the parameters and scans one image.
