Format and Trigger Selection
Binning of the camera pixels can be set in the
Format
drop down menu with the Andor iXon
cameras, but not for the sCMOS pco.edge camera
(Fig. 114). In the
SIM
imaging mode only the
standard (
1004 x 1002 standard;
numbers
indicate pixels) is available. In the
WF
and
Laser
WF
imaging modes you have additionally to the
standard (
1004 x 1002 standard
) the options of
2x binning (
500 x 500 binned 2x2
) and 4x
binning (
248 x 248 binned 4x4
). Selection of
Format
will update the possible
Frame Sizes
.
When selecting the Andor iXon SMLM
camera in the
WF
and
Laser WF
you have
the options to select standard binning (
512 x
512 standard
), 2 x binning (
256 x 256
binned 2x2
) or 4 x binning (
128 x 128 4x4
).
All cameras are run in exclusively in triggered
mode, fre run is not available.
When selecting the sCMOS camera for Localization microscopy in laser WF mode the denoising should be
switched off by selecting
Localization microscopy (denoising off)
in the
Noise Filter
drop down
menu (Fig. 115). The SMLM software will correct the indicidual pixwel noise of the sCMOS camera. For
TIRF and WF applications the entry
TIRF or WF Imaging (denoising on)
should be chosen.
