Processing Options
The
Online Processing Options
tool is only
available in the
Laser WF imaging
mode and
allows to online process SMLM data, control the
laser activation power and activate the holding
focus, all processes that will take place parallel to
data acquisition (Fig. 120).
5.2.9.1
Online Processing SMLM
If the
Online Processing SMLM
checkbox is
ticked, the localization of molecules is computed
and a SMLM image generated on the fly (for 3D-
SMLM you will obtain a localization map only in
2D). You can decide, which kind of fit you want to
use by checking the
Fit Gauss 2D
box to use a full
2D Gauss fit routine, or a
Mask
fit, with a
predefined fixed Gaussian mask, if the box remains
unchecked (Fig. 121).
With the
Peak mask size [pixel]
slider you can
adjust the size of the mask that is used to cut an
identified event out. As a default, the mask has a
size of 9 x 9 pixels. You can alternatively type in
numbers in the input boxes or use the arrowheads
to define the size.
With the
Peak intensity to noise
slider, the noise
threshold is set. An event will be considered as
appropriate, if the standard deviation of its
intensity against the rest image is smaller than the
set value. The higher the number, the more
discriminating the system will look for events. The
default value is 6.
When using
Online Processing SMLM
, a SMLM tab in the image will become available that
contains the SMLM and the sum widefield (SWF) image.
