These microscope functions can also be operated directly on the microscope stand via assigned
buttons or through the touch screen, if available. In that case, any changes are recorded by the
software and displayed in the relevant windows / panels.
For left side port access, the reflector has to be in a
None
position.
If the reflected light lamp is equipped with a software controlled shutter, the
Shutter On/Off
button in
the
Fluorescence
field should be in the
On
position in order to open the shutter. In
Shutter Off
mode,
the shutter is closed and will block the light.
5.1.1
Microscope Control Tool for
Axio Observer
To have access to the microscope functions to
define a configuration, you have to expand the
Microscope Control
tool by pressing the
expansion symbol in the
Microscope Control
tool
tab (Fig. 49).
The
Microscope Control
panel contains the
following functions:
HAL On/Off
button for transmitted light: If the
button is pressed, the control panel appears
(Fig. 50).
If you press the
On
button it will be highlighted
and the Halogen lamp will be switched on
(symbolized by the halo in the lamp symbol).
In parallel the shutter for transmitted light
automatically opens (
On
position in the
Light
control
field and
Open
position in the
Ocular
panel). The light intensity of the HAL lamp can be
varied via the input box or slider.
The light path will be highlighted by color in the
diagram. Press the
On
button again to shut down
the light. The button will go to the
Off
position
and the shutter closes automatically.
The color temperature for photo documentation
can be switched on via the
3200 K
button.
If the
3D
button (only available for systems
equipped with the 3D-SMLM module) is pressed,
the 3D-PALM slider will instantly move in its work
position in the DIC slot of the objective. The button
changes to indicate that the slider is in its active
position. Press the button again to retract the slider
to its load position.
