Setup Manager – Imaging Setup
The
Imaging Setup
tool (Fig. 81) allows
new / existing beam path configurations and track
definitions to be created / edited and saved for the
different imaging modes.
•
Click the
Imaging Setup
tool to open or close
the tool window.
Different imaging modes can be selected by
activating the respective tab.
The following imaging modes are available:
WF
– Widefield imaging using the arc lamp for
widefield illumination and a camera for detection
Apotome
– SIM Apotome mode using laser for
wideield illumination and a camera for detection
Lattice SIM
– Structured Illumination Microscopy
mode using a laser for widefield illumination and a
camera for detection
Laser WF
– Laser widefield imaging including epi-
fluorescence (Epi), Highly Inclined and Laminated
Optical sheet (HILO) and Total Internal Reflection
Fluorescence (TIRF) modes using a laser for
widefield illumination and a camera for detection
The imaging mode is selected by pressing the
appropriate
Imaging mode
button. Below the
Imaging mode
button field the
Acquisition
mode
field allows the selection of the acquisition
mode. For all widefield imaging modes (WF,
Apotome, Lattice SIM, Laser WF) only
Channel
mode is available.
Note that the contents of tool groups and
tool tabs will be specific for a selected
imaging mode and hence change appearance, when imaging modes are switched. Thus it will
depend on the selected imaging mode of the
Imaging Setup
tool, which functions will be
available for other tools.
The
WF
, Apotome, Lattice
SIM
and
Laser WF
light path tools have several information and control
elements in common, whereas others are specific.
