SMLM-Grouping View Control Tab
SMLM-Grouping
is used to assign and group identified peaks to one object (molecule) (Fig. 635). What
peaks should be considered to be derived from one molecule can be defined with the following
parameters:
−
Max On Time
: Maximum number of frames that peaks within the defined capture radius are
allowed to be detected in order to be considered to belong to one molecule. If peaks at the same
location are found in more consecutive frames than all peaks are discarded.
−
Off gap
: Maximum number of frames peaks within the defined capture radius can be missing in
order that the peaks are regarded to belong to the same molecule. If the defined number is
exceeded, than the peaks will be sorted to two molecules and not grouped.
−
Capture radius:
Maximum radius in pixels (as defined by the set rendered localization precision in
x,y and z) within which peaks of consecutive frames must lie in order to be regarded to belong to
the same molecule.
If the
Group
button is pressed peaks are grouped
to one molecule according to the set parameters.
Since peaks are summed, the grouped image will
have fewer objects (Fig. 636).
Since localization precision is encoded by the
brightness of the color, often grouped images have
brighter areas than ungrouped images.
Once grouped the
Ungroup
button becomes
available. Pressing the
Ungroup
button will
accordingly ungroup grouped peaks and the
button will turn inactive again.
An active
Ungroup
button indicates a grouped
SMLM image, an inactive one a non-grouped
SMLM image.
In a multi-channel experiment the channel selection buttons are available. Press
All
to group all channels
with the same setting or press a channel button to set parameters specifically for that channel. Note, all
channels will be grouped according to their actual settings.
If a SMLM drift correction is performed in the
SMLM-Drift
view control tab, a grouped image will
be automatically ungrouped and the
Ungroup
button will turn inactive. To re-group the
Group
button has to be pressed.
The table in the
SMLM-Statistics
view control tab is automatically updated.
