When checking the
Index
box in the
Show
field, the number (index) of the fiducial will be displayed near
the circle.
To toggle between the original drift and corrected drift of a fiducial, press
Clear All
to view original drift,
then followed by
Select All
to view corrected drift wit the
Zoom
box checked (Fig. 612).
Since drift correction is averaged even for one fiducial, drift correction will result in a circular cloud
of positions rather than one position even if only one fiducial is used.
One can also manually select fiducials by pressing the
+
button. The buton then becomes active and
when pointing with the mouse into the SMLM image, a crosshair becomes available as the mouse pointer
(Fig. 611).
Move the crosshair over the position of the assumed fiducial and click into the image with the left mouse
button. The fiducial at the selected position will be added to the table. To inactivate the
+
button, press it
again.
You do not need to have the fiducial exactly in the middle or fully circled. The software looks for
the brightest peak around the crosshair.
All
-user defined-
fiducials are indicated in a yellow color and are indicated as such (Fig. 608). To remove
a fiducial, highlight it in the
Fiducials Table
and press the
-
button.
By pressing the
Drift Table
in the
Show
field the button will be highlighted and the drift table becomes
visible that lists the average drifts of all selected fiducials (Fig. 608). By pressing the button again the drift
table will disappear. The drift table has three columns:

Time
: Frame number in the time series
