Processing with Adjust settings
If
Adjust
is selected the Adjust panel becomes
available offering more options to process the data
(Fig. 383).
Press the triangle of
Advanced Filters
to open to
the
Advanced Filters
field (Fig. 384).
You can choose from the
Parameters
drop down
menu how the parameters should be fitted
(Fig. 385):
−
No Fit
: Theoretical parameters will be taken
and fitting is skipped. This can be done
channel specific and might help avoiding
black planes due to poor raw data, where
the system cannot converge on parameter
values
−
Fast Fit
: Parameters are fitted by using a ROI
of the image that is selected according to
the highest intensity in XY and highest
contrast in z.
−
Best Fit
: Parameters are fitted by using the
whole image.
In the
Advanced Filters
field a
Detrend
check box is available that becomes active
for time series data only. If checked the
algorithm compensates for bleaching or a
decrease in fluorescence over time.
Alternatively you can detrend time series
images before or after processing in the
Detrending
processing tool. This tool allows
in addition to set thresholds above which
detrending should be performed.
With the
Sectioning
slider you can set the
strength to remove the 0-frequencies for the 0
th
order with a pre-defined variance. The higher the
value, the more stringent the filtering will be and
more of the lower frequencies will be removed.
Press the triangle of
Advanced Settings
to open
to the
Advanced Settings
field (Fig. 386).
