You have two options for Processing: Either
Default
or
Adjust
,
which can be chosen from
Parameters
drop down menu (Fig. 372).
Default
uses standard settings, which will work in most of the cases;
Adjust
on the other hand gives access to additional parameters to
be set.
The
Preview
tab shows the
Input
image and a
Preview
of the
output image.
In
SIM
processing the mode of image is displayed in the image mode display area, e. g.
WF
(for
wiedfield),
SIM
Apotome
,
Stripe SIM
and
Lattice SIM
). It is also indicated, if the image was taken with
Leap
mode (
SIM Apotome Leap
,
Lattice SIM Leap
).
By pressing
Apply
the high resolution image will be computed with the current settings.

Processing with Default setting
The tool indicates in the
Processing
drop down menu if the data set
was taken in
2D
or
3D
or
3D Leap
(Fig. 373). For a 2D data set both
3D option are greyed out; for 3D datasets the
3D
or
3D leap
can be
manually changed to
2D
. What is selected will determine how the
data are processed.
When
2D
is selected, every frame is reconstructed with the information of that frame only and the
structuring in the thirds dimension is not used.
If
3D
is selected, the information of the neighboring frames in regard to structuring information and out
of focus contribution will be used in addition to the information of the frame itself. This will result in
better x, y and z resolution and better contrast by removing the out of focus light to a greater extent at
the cost of more computational power requirement.
If
3D leap
is selected, the system will reconstruct 3 planes instead of 1 between the original planes.
Note that
3D
and
3D leap
are are only available, when the image to be processed contains a stack
of >1 frames and
All Z
or
First-Last
was selected as the
Stack processing
mode. Otherwise, the
Processing
drop down menu shows only
2D
and other options are greyed.
If the image was a 2D time series acquired
with
Lattice SIM
, the
Burst Mode
check
box becomes available (Fig. 374). If checked,
the SIM calculation is not any more done by
blocks of 15 phase images (i. e. 1-15, 16-30,
…;
Block mode
), but by 15 phase images in
a gliding or rolling manner with the number
of phase image steps as defined in the
Sliding Windows…phase(s)
slider (
Burst
mode
).
For example, if the slider was set to 1 reconstruction will be performed on phase images 1-15,
2-16,…. Dependent on the slider settings, two events can be resolved to a better extend, however
time resolution is restricted by the 15 images that have to be taken for one reconstructed image.
If a region of interest (
ROI
) is defined in the image, and the
XY ROI
box is checked, then this
ROI
will be exclusively taken for image processing and the rest of the image will not be processed.
