By pressing the
Reset
button all parameters for the selected camera will be reset to the default values.
For SIM and SMLM imaging the default settings of the camera are optimized and should not be
changed. Changing camera parameters in general should be performed with great care.
EMCCD cameras are sensitive to high light exposures. If an EMCCD camera is oversaturated for
extended periods with gain settings above zero, the camera chip will age faster or can even be
damaged. Therefore it is good practice to start always with gain zero and than adapt the gain to
ones need. Also it is good advice to set the gain to zero when in SMLM experiments the bulk of
molecules have to be switched off using high laser powers before they start to blink.
Activate the
Use these settings on restart (not recommended)
check box to start the system with the
set parameters.

Press the
Elyra
tab to open the
Elyra
panel
(Fig. 471).
Tick the
Online pre-processing SR-SIM
check
box to activate pre-processing of SIM data already
during image acquisition. This will save time later
when processing SIM data.
Tick the
Automatic collimator adjustment
check
box to activate automatic adjustment of the
collimator position in dependence of the z-focus
position.
Check the
Use piezo for holding focus
box to
use the
Z-Piezo
stage insert for the holding focus. If not ticked, the microscope focus drive will be used.
It is strongly recommended to use the
Z-Piezo
stage insert for the holding focus as it is preciser and
faster.
Check the
Control holding focus position (only with fiducials)
box, if you want to use
correlation amplitude and PSF width to find the focus of a fiducial using the holding focus. If
unchecked, only the correlation amplitude will be used.
It is recommended to use the
Control holding focus position (only with fiducials)
for the
holding focus with a fiducial as it is preciser and faster. If blinking molecules are used it is better to
deactivate the function.
