If
Peak Tracker
is selected, the peak tracking window will be displayed (Fig. 656).
If peak tracking is active, the mouse cursor changes to a
crosshair within the
Source Image
that allows one to select a
spot by clicking with the left mouse button (Fig. 657), for
example a fiducial, which will be displayed in a 11 x 11 pixel
window. The selected spot is indicated by a cross. During
Online Processing
this would allow to constantly monitor the
fiducial for axial and lateral drift.
The table below the peak tracking image has two or more
columns. The first column is the
Parameter
list, the next
columns are the values of each parameter for each channel
(TV-1, TV-2 etc.). During
Online Processing
these values are
frame wise updated.
The following parameter values are displayed:

Position X [
µ
m]
: Position in
µ
m of the selected spot in
the X direction with the origin at the upper left corner of
the image.

Position Y [
µ
m]
: Position in
µ
m of the selected spot in the Y direction with the origin at the upper
left corner of the image.
−
Number Photons
: Total number of photons of a peak / object.
−
Precision [nm]
: Calculated localization precision of a peak / object in nm.
−
Chi square
:
Χ
2
value of the Gauss fit for the localization precision.
−
PSF width [nm]
: Width of the point spread function (PSF) calculated from the Gauss fit.
−
Background variance
: Variance of the number of background photons.
Below the table are also listed the following two values:
−
molecules in current frame
: peak counts per frame
−
molecules in all frames
: accumulated peak counts to the present frame
These two values are, however, updated only every 1000th frame.
