The
Detrending Processing
function (Fig. 441)
can be used to compensate for fluorescence loss
during time lapse experiments that might be
induced by bleaching.
Select the image you want to detrend by pressing
the Select button. Note, if the image is not a time
series, the
Select
button stays inactive.
Choose how you want to detrend the data. If the
Refer to Previous
checkbox is ticked, the
software detrends the data of a frame in reference
to the previous frame (Fig. 442). If the checkbox is
not ticked, the software detrends the data in
reference to the last image compared to the first
image by linear interpolation.
If the
Use XY ROI
check box isticked and a ROI
selected, detrendingwill be derived from the ROI
but applied to the whole image.
You vcan input a threshold in the
Threshold
input
box defining the intensity above which detrending
occurs. This allows to avoid if signal is well
abovebackground noise to raise background as
well if the threshold is set above the background
value.
