In the following two procedures are given to adjust the level of the stage insert. Procedure 1 uses epi-
fluorescence light, procedure 2 reflected light.
For epi-fluorescence light either the X-Cite lamp or laser light can be used. Using the X-Cite
lamp, if available, is recommended, since the incubator can be open and adjustment with the
adjustment screws can be done under observation. In case of laser light the incubator has to be
closed and adjustment cannot be done simultaneously with observation. Instead, having done
an adjustment, the incubator has to be closed to monitor the outcome and the adjustment has
to be done iteratively.
The level-adjustable stage insert can become helpful when working with water objectives as
their PSFs are sensitive to tilt in the sample.

1.
Take a fluorescent sample like embedded
beads and mount centrally over the objective.
(Note: you can also use your own fluorescent
sample). Use WF illumination with the X-Cite
or laser light and focus manually onto the
sample using the stand’s focus drive (Fig. 7/
A
).
2.
Set stage to zero by pressing the
Set Zero
button in the
Stage
tool (Fig. 8). Set focus to
zero by pressing the
Manual
button in the
Focus
tool (Fig. 9).
