Manual
•
For manual processing, deactivate the
Auto
checkbox (Fig. 254).
•
In the preview window you can check again
whether the right image type is selected.
Note that this preview will not show a full
SR processing result.
•
Use the strength slider to get a stronger or weaker
result as with the auto processing. You might use
the same values as used in the 2D preview
processing in the
Airyscan
tab.
•
Press the
Apply
button at the top of the dialog.
Processing will start and might take a few minutes.
A new image will open with the result of the manual
processing (Fig. 254). All images in a time series or
Z-Stack will be processed.
Note that with 3D data you have the choice of
a 2D or a 3D processing by selecting the
method from the dropdown list (Fig. 255). A 3D
processing will take longer but will result in a
higher resolution increase since also the axial
Airy data will be used for the processing.
Use the same approach for R-S mode. However
the achievable resolution will not match the
resolution of the SR mode as the sampling in
XY is not sufficient for superresolution. The
images will still show higher resolution and a
considerable increase in signal to noise ratio.
VP data processing
In case a VP mode image was selected, the manual
processing slider will function for virtual pinhole
parameter adjustment.
This allows to adjust the confocal sectioning after
acquisition by binning the respective airy rings
together.
•
Press the
Apply
button at the top of the dialog.
Processing will start and might take a few seconds.
A new image will open with the result of the selected
sectioning. All images in a time series or Z-Stack will
be processed.
