Acquisition Parameter – Shuttle and Find
Shuttle and Find
allows calibration and imaging of sample positions in laser wide field (laser WF) and
SIM mode for reproducible repositioning after transfer to scanning electron microscopes (SEM).
For
Shuttle and Find
different types of sample holders are pre-designed. In addition the user can define
other correlative sample holders.
The correlative sample holders have three fiducial markers enabling a three point calibration.
The calibration can be done in transmitted light or reflected light.
A motorized microscope stage is mandatory.
The workflow is unidirectional from SR (laser WF, SIM) to SEM only.
Different types of sample holders are available
(Fig. 175), e.g.:

Life Science cover glass 22x22
−
Life Science for TEM Grids
−
MAT Universal A
−
etc.
The Life Science cover glass 22x22 or the
sample holder for TEM Grids should not be
used together with Axio Imager and
Axio Examiner equipped with a level
adjustable sample holder. During the
calibration procedure the objective or
neighbouring objectives will collide with the
level adjustable holder.
Creating of user-defined correlative holders
In the calibration dialog window new correlative
holder files can be defined (Fig. 176).

Click on the
+
button in the
Select Template
dialog.
•
Type in a name for the new holder / sample
carrier. An image of the new holder can be
loaded.
In the holder file the distances (in millimeters)
between the first and the second marker and
between the second and third marker have to be
inserted.
The distances can be determined using the
Positions
button in the
Dimensions
control view and the
functionality for
Marks
in the
Stage
tool in the Left Tool Area.
•
Activate the
Positions
button and click on the first correlative marker.
•
Determine the coordinates by clicking on
Mark
in the
Stage
tool.
