Shading Correction
Shading Correction is a tool to manipulate images
which are unevenly illuminated over the field of
view. This works with single images, time series, Z-
Stacks, or tiled images. Click
Shading
correction
to open the interface for using that function
(Fig. 440).
Select
the image (or image series, tiled image, or
Z-Stack) for which the shading correction should
be applied by clicking
Select
next to
Input image
.
When clicking
Apply
the system calculates a
reference image from the input image(s) which is
used to correct the input image(s). The corrected
image is displayed in the center screen area.
In addition, the calculated
reference
image
can
be stored and used for further shading corrections
of other images. Therefore the check box in the
lower part of the tool box must be checked.
Specify the folder where the image should be
stored in the text box next to
Path
.
Browse
opens
the windows folders from which you can select. A
suggestion for the name of the image is made
based on the original name of the image. Change
this if needed.
This reference image can then be loaded and
selected as a reference image for the shading
correction of subsequent images, which are then
processed much quicker.
The reference image must be taken from an image
that was acquired with identical settings for
objective, zoom, image format, and orientation.
