Pay attention to the following guidelines for the Experiment Designer

Each Block generates a separate image file. Loops going back to a Block will add the newly
acquired images to the already existing file of this Block. Hence, only one image per existing block
irregardles of the set loops and repetitions will be generated. The time stamps of the final image
represent the acquisition time of each image with the first image set to time point zero.

Auto Save settings apply to the complete Multi Block experiment. Auto Save however produces a
separate image for each Block and for each repetition of a Block. It serves as a back up. Images
should therefore always be saved once the experiment is completed to save the data as one image
file per Block.

Image files are automatically named consecutively keeping the number for the image constant for a
given Multi Block Experiment. The Block number always starts with 1 and the number is
consecutive within the experiment.

Use the Macro "ModifySeriesZEN.lvb" to combine auto-saved images into consecutive time series
images if needed. See Chapter 2: MACROS AND VISUAL BASIC for further explanations.

Set desired stage position(s) for each new block separately using the Positions Tool. For multi-
position experiments in combination with Z-Stack acquisition the Z coordinate from the Position list
is referring to the center position of the Z-Stack.

Bleaching experiments are restricted to one position per Block. If more than one position is defined
for a Block, Bleaching is disabled.

Positions and Regions are not saved with the experiment, only with the image. Positions cannot be
reused from the resulting image but have to be stored as separate position list(s). See also
section 5.2.21.1 on using position lists.

Autofocus Strategies use the center position of a Z-Stack as the reference position. For a given
Z-Stack go to the center position and if necessary, click Center within the Center tab of the Z-Stack
tool once to confirm the reference.

Tile scan settings (number of tiles and % of overlap) are saved with the experiment. Settings within
Bounding Grid and Convex Hull are not saved.

All imaging modes defined in the Imaging Setup Tool (i.e. WF, SIM) can be assigned to different
Blocks and used in mixed mode experiments.
