7.4.7 Polarising a (polarographic) METTLER pO
2
Sensor
Polarographic pO
2
sensors from the manufacturer METTLER must
be polarised at initial operation or after disconnection from the volt-
age source. Correct calibration is not possible otherwise. This
means that the pO
2
sensor must be polarised before calibration.
NOTICE!
Details about polarisation, calibration, use mainte-
nance and servicing can be found in the separate
sensor documentation of the manufacturer.
7.4.8 Calibrating the pO
2
Sensor
The bioreactor system is preconfigured either for using polaro-
graphic METTLER pO
2
sensors or HAMILTON pO
2
sensors of the
type Visiferm DO ARC.
Generally it applies that: calibrating the pO
2
sensor should be per-
formed after sterilisation because sterilisation can change the gra-
dient of the pO
2
sensor.
A 1-point calibration to 100 % is generally sufficient for an exact
measurement and should be carried out again before each fermen-
tation.
The zero point (0 % calibration) should be checked at regular inter-
vals of about 6 months.
A detailed description how to calibrate the pO2 sensor can be
found in the separate touch screen software manual from the
equipment manufacturer.
METTLER pO
2
sensors are polarographic and therefore need to be
polarised before calibration.
NOTICE!
Detailed information on the calibration, use,
maintenance and servicing can be found in the
separate sensor documentation of the manufac-
turer.
