7.2.3 Preparing the Reagent Bottles, Hoses and Pumps
WARNING!
Risk of injury due to bursting of corrective rea-
gent hoses!
Reagent hoses can burst or come off the addition
port adapters during sterilisation in the autoclave
due to overpressure or damaged hoses. Spurting
out of or spilled hot and/or corrosive corrective rea-
gent may cause severe injuries to the user when
taking the culture vessel out of the autoclave.
Therefore:
– Only sterilise reagent bottles with hoses that are
intact, thoroughly rinsed, empty or filled with wa-
ter - never sterilise hoses filled with corrective
reagents!
– Always secure reagent hoses with cable ties.
– Only sterilise reagent bottles that are equipped
with a short and open hose line, fitted with a
clean and dry filter for pressure equalisation.
– Always clamp off reagent hoses during sterilisa-
tion.
CAUTION!
Risk of loss of property due to usage of highly
corrosive hydrochloric acid!
Usage of the highly corrosive hydrochloric acid HCl
as corrective reagent leads to damages to compo-
nents made of stainless steel such as e.g. vessel
top plate and inlet ports.
Therefore:
– Do not use hydrochloric acid (HCl) as corrective
reagent
– Use non-corrosive acids, e.g. phosphoric acid,
instead
7.2.4 Calibrating the Pumps
Not calibrated pumps show the duration in seconds and the num-
ber of rotations. The pumps must be calibrated to show the deliv-
ered volume. Calibration must be done before sterilisation.
