Sterilising the vessel with culture medium method
WARNNG!
Risk of injury due to splashing liquids when
taking a vessel out of the autoclave which is
too hot!
Superheating and subsequent boiling may occur
when sterilising liquids. Liquids may suddenly
splash out when taking a vessel out of the auto-
clave filled with culture medium at too high a tem-
perature (>80
°
C).
Therefore:
– Sterilise a vessel in the autoclave only filled with
culture medium when using an external refer-
ence sensor (fits into pocket in the top plate).
– Never take the vessel out of the autoclave at a
temperature higher than 80
°
C.
Fill the vessel with the culture medium.
Sterilise the vessel.
Add sterile water and any heat-labile components under ster-
ile conditions.

This method is often used for bacterial cultures
Sterilising the vessel without culture medium method
WARNING!
Contamination risk due to insufficient sterilisa-
tion result!
Development of steam is not possible when sterilis-
ing a completely empty and dry vessel. This may
lead to insufficient sterilisation results and carries a
contamination risk!
Therefore:
– Always fill the vessel with approx. 10 mL per li-
tre of working volume of water, when using the
sterilisation method “sterilising the vessel with-
out medium”.
