Adjustment
Once the lamp is installed (and after a 24-hour burn-in period), it may need an adjustment to
maximize the sensitivity of the Fluorolog
®
-3. To do this, Jobin Yvon Inc. recommends run-
ning a water Raman scan to check wavelength accuracy, and then monitoring the peak in-
tensity while adjusting the vertical height of the lamp:

Insert a water-filled cuvette in the sample
compartment.

Take a water Raman scan with the following
parameters:
Excitation wavelength
350 nm
Scan range
365 nm to 450 nm
Increment
0.5 nm
Integration time
1.0 s
1.25 mm (single-grating spectrometer)
Slits
2.50 mm (double-grating spectrometer)
Acquisition mode
