The Site Map in the upper right represents the number of sites in each well and the position of
the sites to acquire for each well. Use the Site Map to control some of the movable physical
components of the ImageXpress Confocal HT.ai system and to configure the sites to acquire.
Sites you select to acquire display green and sites you choose to not acquire display gray.

Below the Plate Map and the Site Map, wavelength tools provide shortcuts to various functions
related to the acquisition wavelength. Each icon performs the action at the well where the
stage is currently located and in the wavelength that currently displays in the Active
Wavelength field.
The
WI (Water Immersion) Status icon displays when your instrument has the Water
Immersion option and you select a water immersion objective. Click the icon to display the
Water Immersion Control dialog which provides the status of the water immersion system
and basic water immersion maintenance steps. See
Water Immersion Option on page 179
.
Use the Active Wavelength drop-down to select an acquisition wavelength.
Use the
Snap icon to acquire an image without performing (or re-performing) an
autofocus.
Use the
Start Live icon to start and stop Live mode.
WARNING!
The external light source for the ImageXpress Confocal HT.ai system
is a Class 4 high-power laser product. Prolonged exposure can cause plate
materials to smolder or burn. If live-mode exposure is longer than 30 seconds, the
software automatically closes the laser shutter.
CAUTION! Using Live mode for extended periods of time increases the risk of
photo bleaching the site.
Use the
Focus icon to perform the Autofocus routine and then acquire an image.
Use the
Test icon to perform the Autofocus routine and then acquire a Z series and
present the corresponding 2D projection when you set up a z acquisition.
Use the
Preview icon to perform the Autofocus routine and then acquire a Z series at
each of the wavelengths you set up in the protocol and present the corresponding 2D
projection when you set up a Z acquisition.
Note: There are a few cases where configuring a plate acquisition protocol with the
Active Wavelength is applicable. These cases are discussed where appropriate in this
section. These tools are primarily used when you set up plate acquisition.
