3. To navigate the Z Series for a site or well position and modify the coordinates for the Z
motor before you acquire any images, do the following:

Drag the Ruler Zoom slider (below the Z series diagram) or use the mouse scroll wheel
to zoom in or zoom out on the diagram.
Focus displays on the Z series diagram to indicate the position of the Z motor after
autofocusing and wavelength offsets are applied. Do the following to modify the Z
stack acquisition settings:
Click the Focus active wavelength tool to place the Z motor at the Focus indicator
in the Z series diagram and display its current position in µm. The ruler markings
indicate the distance in µm above and below the Focus position.
Drag the Top flag or the Bottom flag to set the top and bottom positions for the Z
series acquisition relative to the Focus position. As you adjust these values, the
software automatically calculates the values for the # of Steps field and the Range
field. The position in µm to which you move a flag temporarily displays on the flag.
Select the Center Z Series Around Focus Result check box to evenly space the Z
series planes above and below the Focus position. When you drag a Top or Bottom
flag on the Z series diagram to set the position for the Z series acquisition, the
software automatically moves the other position by the same total distance in µm.
Clear the Center Z Series Around Focus Result check box and click Set B to designate
the bottom Z plane to be acquired at each site.
