Using Oil-Immersion Objectives
You can use oil-immersion objectives with the ImageXpress Confocal HT.ai system for
research-mode imaging. For ordering information, contact your sales representative.
Note: We do not recommend using an oil-immersion objective to scan an entire plate.
To apply oil to the objective:
1. In the MetaXpress software, from the IX Taskbar, click Adjust Correction Collar to access
the specific objective. If prompted to select the mode of access, select From the Top.
2. When prompted, remove any plates in the instrument.
3. Use a dropper bottle to sparingly add oil to the top of the objective.
4. Insert the sample with either a thin glass coverslip, or in a plate with a thin glass bottom,
and then click Continue.
Note: Oil-immersion objectives are not compatible with plastic plates.
5. In the Plate Acquisition Setup dialog, select the Configure tab then select the Acquisition
tab.
6. Select the Enable Image-Based Focusing check box.
Note: Only use Image-Based Focusing with oil-immersion objectives.
7. Select the Autofocus tab.
8. Click the Initial Well for Finding Sample drop-down and select Skip Find Sample.
Note: Oil-immersion objectives are not compatible with the Laser Autofocus
Wizard.
9. Slowly step up the objective manually until you are near to focus.
10. When you are done with the oil-immersion objective, click Adjust Correction Collar to
access the objective again, and use lens paper to clean the top of the objective. If
needed, remove the objective for better cleaning. See
Cleaning Objectives on page 211 .
