Transmitted Light Phase Ring Alignment
The Molecular Devices Field Service Engineer performs phase ring alignment when installing
the ImageXpress Transmitted Light option. After the phase ring is aligned, the microscope
generally holds its position for objective changes. Phase ring alignment should be checked
periodically, especially when you see degradation of phase contrast image quality.
The phase ring alignment procedure requires a special lens to be placed in position before
starting the phase ring adjustment process.
On the ImageXpress Confocal HT.ai system, you must install the phase ring adjustment lens in
Position 8 of the emission wheel to perform the phase ring adjustment process. After the
adjustment process, you must remove the phase ring adjustment lens and replace it with the
original filter.
Installing the Phase Ring Adjustment Lens
To install the phase ring adjustment lens:
1. Exit the MetaXpress software.
2. Remove the emission filter currently installed in Position 8 of the emission wheel. See
Removing Emission Filters on page 201
for details.
3. Store the filter in a safe place protected by a non-scratch paper, such as lens paper.
4. Install the phase ring adjustment lens in Position 8 of the emission wheel. See
for details.
5. Restart the MetaXpress software.
Using the Phase Ring Adjustment Lens
To use the phase ring adjustment lens :
1. In MetaXpress software, do one the following to display the Device Control dialog:
From the simplified menu, click Control > Devices > Device Control.
From the standard menu, click Devices > Device Control.
2. Click Component Control to display the Compound Control dialog.
3. Click the Component drop-down and select the IXConfocal Module Emission Wheel.
4. Click the Position drop-down and select Position 8.
5. Click Close to display the Device Control dialog.
6. Click Close.
Aligning the Phase Ring
Note: Confirm that the instrument is in widefield mode before aligning the phase ring.
To align the phase ring:
1. Confirm that the phase ring adjustment lens is in place.
The phase ring adjustment lens is configured in the emission wheel Position 8 and is only
used during the phase ring adjustment process using the MetaXpress software. We
recommend leaving the phase ring adjustment lens installed in the emission wheel
Position 8.
