12. Adjust the position of the Z-stage to obtain a reasonably sharp image of the objective
phase ring.

13. Use the region tools in the software to mark the position of the phase ring. Use the
rectangular region tool to make sure that you capture both horizontal and vertical axes.

14. Do one the following to save the marked positions:
From the simplified menu, click Measure > Regions > Save Region.
From the standard menu, click Regions > Save Regions.
15. Manually rotate the condenser turret to the annulus for the phase objective (PhL for 4x, Ph1
for 10x and 20x, Ph2 for 40x).
The annulus is at the front of the condenser turret.
Phase contrast objectives typically have their specifications indicated in green letters on
the outer barrel. These specifications also include the matching annulus designation (for
example, PhL, Ph1, Ph2).
