Find the focal plane, then use
Go within range
and
+ -Range
to set how many images the
Scanner should take in the Z-direction. (The distance between the two images in Z-
direction is 0.2 μm). Under the
Output folder
field, select the folder where the images are
saved (it is a good idea to create a new folder for each recorded image sequence), and
use the
Prefix of output filename
option to add a prefix to each image in the sequence.
Once you have finished, click
Generate PSF
button and the Scanner will start recording
the image sequence.

As the last step of setting the deconvolution process, load generated PSF results into the
software. Select the appropriate PSF output for each of the filters in
Settings
mode under
the
Browse
tab of the
PSF
settings. Enter the number of images in the series that the PSF
consist of, then select the folder under
Folder name
into which the PSF results are saved.
Click
Save
to accept modifications.
6.
Turn on deconvolution function by selecting
D
on the
Filter settings
panel in
Microscope
mode. This way scanning can be performed with the correct parameter settings.
Notice:
Deconvolution can be performed in Extended focus or Z-Stack scanning modes only.
