Performing Experiments with the Water Immersion Option
The process for acquiring images with water immersion objectives is essentially the same as
the process for acquiring images with air objectives. This section describes the additional
steps for using water immersion objectives:

Before you acquire images with a water immersion objective, ensure that:
The source bottle has enough water for the experiment.
The waste bottle has enough space for the water that will be drained from the water
immersion objective.
The bottle caps are screwed on tightly.
To refill or empty a water bottle:
1. Unscrew the bottle cap and, while holding the cap and tubing, remove the bottle from the
water immersion controller.
2. Drape the tubing and cap over the front of the water immersion controller (to prevent the
tubing and cap from coming into contact with the table surface or other potential
contaminants).
3. Fill or empty the bottle.
CAUTION! Use only distilled water in the source water bottle. Deionized water can
be used, but is not required. Do not use tap water and do not reuse the water that is
in the waste bottle.
Note: When filling the source bottle, add water slowly. Adding water too quickly
can cause air bubbles to form.
4. Insert the tubing into the bottle and then put the bottle back into the controller.
5. Tighten the bottle cap.
6. In the software:
a. If the Plate Acquisition Setup dialog is not already open, click Screening > Acquisition
Setup.
b. In the Plate Acquisition Setup dialog, click WI Status.
The Water Immersion Control dialog opens.
c. Click Refill/Empty Bottle.
The Bottle Maintenance dialog opens.
d. Click Test Vacuum and Test Pressure to ensure the bottles are properly sealed.
The water immersion system has multiple leak detectors and pressure sensors to determine if
there is unintended water loss. When a sensor is triggered, the system automatically shuts off
the pump and closes the valves to prevent damage to the instrument. For more information,
