2. Configure the time series acquisition options.
Option Description
Number of
Timepoints Specify the total number of time points to acquire. If you change this value, the
software calculates the Number of Timepoints and the Interval to update the
Duration field.
Perform
Time Series
For Available when the Number of Timepoints is greater than 1. Specify the order for
the software to use to acquire images at multiple time points and defines the
time interval and total length of the time lapse acquisition routine. Select one of
the following:
One Well Then The Next acquires a set of wavelength images at each site in
the well at each time point.
Note: This option is most common with a photoactivation event.
One Row Then The Next collects all images that are in one row of wells at
each time point. After the software collects the series, the next row is
acquired.
Note: This option requires a longer time interval because the
software acquires all wells in a row.
One Column Then The Next collects all the images in one column of wells at
each time point. After the software collects the series, the next column is
acquired.
Note: This option requires a longer time interval because the
software acquires all wells in a column.
All Selected Wells acquires all wells at each time point in the acquisition
routine.
Note: This option requires a longer time interval because the
software acquires all wells in the plate.

Note: The Timelapse Autofocus setting on the Autofocus tab determines any
refocusing the software does after the images for the first time point are acquired.
