Verifying the Plate Reference Point (A1 Center)
This procedure requires the metal slide holder plate that ships with the instrument to verify that
the plate reference point is in the center of well A1.
To verify the plate reference point is properly set:
1. In the MetaXpress software, do one of the following to display the Plate Acquisition Setup
dialog:
From the simplified menu, click Screening > Acquisition Setup.
From the standard menu, click Screening > Plate Acquisition Setup.
2. Select the Configure tab.
3. On the Configure tab, select the Objective and Camera tab.
4. Click the Magnification drop-down and select the lowest power objective (max. 10x).
5. In the Camera Binning field, enter 1.
6. Click the Acquisition Mode drop-down and select Widefield.
7. Select the Plate tab.
8. Click the Plate Name drop-down and select 96 Wells (8x12).
9. Select the Sites to Visit tab.
10. Under Site Options, select Single site.
11. Select the W1 tab below the Acquisition > Wavelengths tab.
12. Click the Illumination drop-down and select FITC.
Note: Filters other than FITC are acceptable, but the contrast might not be as high
as with the FITC. Exposure times vary significantly depending on the light source
and filter choice.
13. In the Exposure field, enter 100 milliseconds (ms).
14. Click Screening > Plate Acquisition and Control to display the Plate Acquisition and
Control dialog.

15. Click the Wavelength drop-down and select W1 - FITC.
16. In the Step Size field, enter 250.
17. Click Eject Plate to open the instrument top door.
