Adjusting Objective Correction Collars
The ELWD (extra long working distance) Nikon objectives and the water immersion objectives
have an adjustable correction collar that minimize spherical aberration in the image of the
specimen. The collar adjustment range varies depending on the objective. Change the range
setting to adjusts the distances between components inside the objective barrel. Image
quality and resolution are very much dependent on properly setting the objective collar.
The setting depends on the thickness of the plate well or slide on which you mount the
specimen. In general, set the correction collar for the physical thickness of the plate or slide
that you image.
Determine the physical thickness in one of the following ways:
Get the plate specifications from the plate manufacturer.
Break a spare plate and use calipers to measure the thickness.
Measure the optical thickness with the laser autofocus and multiply it by the refractive
index of 1.59 for polystyrene or 1.52 for glass.
After you determine the thickness of the plate or slide, you can adjust the correction collar.
CAUTION! If the thickness of the plate, slide, or cover slip is out of the range of the
correction collar, you should not use it with the objective.
Before you adjust the correction collar on an objective, read and follow the Maintenance
Precautions on page 189 .
Tip: You might need a flashlight to view the markings for the graduated scale on the
barrel and its current setting.
To adjust an objective correction collar:
1. Start the MetaXpress software.
2. In the IX Taskbar, click Adjust Correction Collar to access the objective.
3. If the objective is accessible from the side, open the upper, hinged side panel door of the
instrument.
4. If the objective is in one of the center positions, access it from the top.
When prompted by the software, remove the plate or slide holder, and then click
Continue.
5. Locate the correction collar on the objective.
Loosen or remove an air objective to locate the correction collar, if needed.
CAUTION! You must contact a Molecular Devices Field Service Engineer to remove
a water immersion objective.
6. Rotate the correction collar to its new setting. If needed, replace or tighten the air
objective.
7. If you access the objective from the side, close the upper, hinged side panel door.
8. Click Continue.
9. If you access the objective from the top, then replace the plate or slide holder when
prompted, and then click Continue.
10. Test the correction collar setting by examining the image quality of acquired images.
If the quality has degraded, repeat these steps to re-adjust the correction collar.
