Verifying the Laser Autofocus Sensor
This procedure uses a bead plate to verify that the laser autofocus sensor responds.
To verify that the Laser Autofocus sensor responds:
1. In the MetaXpress software, do one of the following to display the Plate Acquisition Setup
dialog:
From the simplified menu, click Screening > Acquisition Setup.
From the standard menu, click Screening > Plate Acquisition Setup.
2. Select the Configure tab.
3. On the Configure tab, select the Acquisition tab.
4. Select the Enable Laser-Based Focusing check box.
5. Select the Objective and Camera tab.
6. Click the Magnification drop-down and select 10x.
7. Select the Plate tab.
8. Click the Plate Name drop-down and select Bead Plate IXM-C.
9. Click Eject Plate.
10. Load the Tetra Speck bead plate and click Load Plate.
11. On the image of the plate, right-click well A1 to move the stage to that well.
12. On the Configure tab, select the Autofocus tab.
13. Click Configure Laser Settings to display the Configure Laser Autofocus Settings dialog.
14. Click Preview Pass to display a Preview Pass image window with a graph of focus
intensities compared to the Z-position. You want to see graphs of sharp peaks in red and
green.
