Configuring Adaptive Acquisition for Well Sites
The Sites to Visit tab Adaptive Acquisition option is a computational algorithm that counts the
number of cells on the fly during sample acquisition. This increases the chances of collecting
valid data in every well. The number of sites the software acquires per well is based on the
number of cells per well.
Note: Adaptive acquisition can significantly reduce acquisition time for multi-
wavelength acquisition that requires a minimum number of cells per well or for samples
with differing conditions across the plate.

To configure an adaptive acquisition:
1. Select the Sites to Visit tab.
2. Select the Adaptive Acquisition Site Option.
3. Select the Custom Field of View (%) check box to specify a percentage of the field of view
for the camera to acquire, and then enter the X (width) value and Y (height) value. This is
useful to acquire images from an unevenly illuminated full field or to acquire images that
include the area outside the well boundary.
Note:
The full field of view for the camera is 100 percent (100% in the X field and 100%
in the Y field). For example, if the full field of view width is 1000 µm, then to
acquire a width of 500 µm, enter 50 in the X field.
Custom field of view is compatible with full-size shading correction images
when you use Legacy Correction. See

4. In the Columns field and the Rows field, enter the maximum number of sites to visit. For
example, Columns = 2 and Rows = 2 acquires up to four sites, Columns = 3 and Rows = 3
acquires up to nine sites.
Note: Maximum allowable values: Columns 45 and Rows 35.
