Confirming Laser Autofocus Settings for Plate Files
Before you use a plate file, confirm that the laser autofocus settings are optimized for the plate.
Note: If you use an objective with a correction collar, ensure that the correction collar is
set appropriately for the plate. See
Adjusting Objective Correction Collars on page 213
.
The laser autofocus system moves the vertical (Z-axis) position of the objective to a point
where the bottom of the well is in focus before the software acquires each site during plate
acquisition.
Note: The Laser Autofocus Wizard calculates measurements as accurately as possible.
Some manual verification and adjustment of the settings might be necessary to
optimize the results.
To confirm the laser autofocus settings:
1. Prepare the plate to test by putting water or buffer in several of the wells.
2. In the MetaXpress software, do one of the following to display the Plate Acquisition Setup
dialog:
From the simplified menu, click Screening > Acquisition Setup.
From the standard menu, click Screening > Plate Acquisition Setup.
3. Click Eject Plate.
4. Place the test plate in position and then click Load Plate.
5. Select the Configure tab.
6. On the Configure tab, select the Plates tab.
7. Click the Plate Name drop-down and select the plate for which to configure laser settings.
8. On the Objective and Camera tab, select a magnification.
9. On the Acquisition tab, select the Enable Laser-Based Focusing check box.
10. Use the navigation tools at the top of the Plate Acquisition Setup dialog to move to an
appropriate well and site.
11. On the Autofocus tab, click Configure Laser Settings to display the Configure Autofocus
Settings dialog.
Note: If the Configure Laser Settings button is red, the autofocus was not
configured for this plate and objective configuration. To correct this, run the Laser
Autofocus Wizard.
12. Click Find Sample to display the Plate Acquisition report. Verify that the focus status
reports Focus Found and that the resulting focus position is appropriate for the plate.
Note: If the focus status reports Focus Not Found, or if the Fine z-position result is
wrong, use the Laser Autofocus Wizard to optimize the laser autofocus settings.
13. Click OK in the Plate Acquisition report.
14. Click Close in the Configure Laser Autofocus Settings dialog.
