Troubleshooting Water Immersion Issues

If you encounter an issue with the water immersion system, try performing one or more of the
procedures below before contacting Molecular Devices.

If the WI Status icon

is not displayed on the Plate Acquisition Setup dialog, verify in the

Meta Imaging Series Administrator application that you are using the correct hardware profile.

If the system is preventing plate acquisition, confirm that the plate settings are accurate and
that the plate is compatible with the objective.

If the system is unable to focus on the plate, follow these steps:

1. Review the plate settings.

2. Visually inspect the water bolus to ensure that it is formed properly on top of the objective.

3. Run the Laser Autofocus Wizard.

If a message indicates that a leak has been detected, follow these steps:

Note: To access leak recovery instructions in the software, on the Plate Acquisition

Setup dialog, click WI Status

to display the Water Immersion Control dialog and

then click Leak Recovery Instructions.

1. Eject the plate.

2. Move the stage so that you can see the objective.

3. Use a lens cleaning tissue to soak up water that is on the top and sides of the water

immersion objective. Look for water that may have gone over the edge of the objective or
elsewhere in the system; dry any water found, provided that you can reach it safely.

4. Check the water immersion tubing to ensure it is not pinched or kinked.

5. Tighten the bottle caps. If the Leak Recovery Instructions dialog is not already open in the

software, on the Plate Acquisition Setup dialog, click

WI Status to display the Water

Immersion Control dialog.

a. To test the vacuum seal, click Test Vacuum.

b. To test that the vacuum line is not clogged, click Test Vacuum Line.

c. To test that water is dispensed correctly, click Test Dispense.

If a message continues to indicate an error or issues persist, contact Molecular Devices.
