To clean the flowcell with dilute nitric acid, do the following:
1.
Remove the flowcell assembly from the detector housing
before cleaning with a nitric acid solution. This will prevent
possible leaks from harming the mechanical and electronic
components of the detector.
2.
Flush the flowcell with water before proceeding. This step is
very important!
3.
Prepare a 20 % (v/v) solution of nitric acid in HPLC-grade
water.
4.
Pump the nitric acid solution through the flowcell with the
chromatographic pump.
5.
When using an LC pump to flush the flowcell, be sure to
replace your column with tubing and make sure water was the
last solvent in the pump and solvent reservoir. If you use a
syringe, always draw the solution
through
the flowcell.
6.
After you have finished the cleaning procedure and before
returning to the buffer solution and chromatographic solvents,
pump another 25 to 40 mL of water through the flowcell to
remove all traces of nitric acid. The pH of the outlet stream of
the flowcell can be monitored to ensure that all of the acid has
been completely flushed out.
7.
Reinstall the flowcell assembly.
INSTALLING THE
FLOWCELL
To install the flowcell, do the following:
1.
Connect the inlet and outlet tubing to the flowcell.
2.
With the inlet tube on the bottom left and the flowcell slot on
top, slide the flowcell assembly into the detector under the
alignment pin, slide the retaining block over the alignment pin,
and hand-tighten securely in place with the retaining block
knob.
3.
Connect the inlet tubing to the chromatographic column and
the outlet tubing to the waste reservoir.
4.
Guide the tubing through the slots in the flowcell compartment
cover and secure the cover by pushing in on the snap latch at
the top of the cover.
5.
Connect the power cord to the rear panel of the detector.

Use the Wavelength Calibration and Dark Current Calibration
procedures outlined in Chapter 5.
