10. On the eighth and final page of the wavelength calibration
wizard, the Finish button is enabled after the new wavelength
settings are downloaded to the detector. Click on the Finish
button to complete the wavelength calibration procedure.
PERFORMING AN
ARRAY
CALIBRATION
FROM ATLAS
To perform an array calibration from the Atlas data system, do the
following:
1. Prepare the detector for a wavelength calibration:
a. Replace the column with a flow restrictor.
b. Start pumping HPLC-grade water through the flowcell at a
flow rate of 1 mL/min.
c. Turn on both lamps and wait for one hour to allow the
temperature of the detector to equilibrate. The dark current
that is compensated for by performing an array calibration
is a function of temperature.
2. In the Array Calibration group box, click on the New
Calibration button to launch the Array Calibration wizard.
3. In the first page of the wizard, click on the Next button to
proceed.
4. In the second page of the wizard, the following steps are
performed as indicated:
Checking lamp states
Retrieving current calibration
Shutting off lamps
Getting calibration data
Restoring lamp states
Retrieving current method
Downloading calibration method
Waiting for equilibration (5 sec)
Restoring saved method
Calibration complete
The Next button is enabled after the program completes the
steps listed above. Click on the Next button to proceed to the
third page of the wizard.
5. On the third page of the wizard, click on the Apply button to
accept the calibration.
6. Click on the Finish button to exit the wizard.
