To define the size of the sample embedding
cylinder, you can adjust its diameter. Use the drop-
down list at the top of the
Specimen Navigator
tool window next to
Specimen diameter
. You
can pick default values or manually insert a
diameter. Recently used specified diameters are
listed next to
Recent
(Fig. 43). The diameter
specified is directly influencing the
Collision
Check
(see
3.4.3.3 System Options – Hardware
and should match the actually used sample.
For an exchange of a sample holder click the
Load
Position
button. The same holder will be driven to
the upper most position. Now an easy access of
the sample holder through the upper system cavity
door is possible.
You can define a position which you might want to return to during the course of imaging by pressing
the
Set Home Position
button after putting the sample to the desired spot. To return to this position
you click the
Home Position
button.
The distance between the Load Position and the y-position for imaging is often quite large. This results in
high numbers for the y-coordinate which are not easy to handle. The current y-position is set to zero
when you press the
Reset Y Reference
button.
This will not affect the coordinates in the
Multiview-Setup
tool window (see section
Multidimensional Acquisition – Multiview-Setup
). Here the y axis parameters will retain their original
naming and need no changing.
3.1.4
Incubator Tool
To have access to incubator functions for defining the atmosphere and temperature, expand the
Incubator
panel by pressing the triangle in the
Incubator tool
tab.
All incubation components you wish to use during the imaging session must be connected to the
Lightsheet Z.1 system and turned on before starting the ZEN software. Once ZEN is running you
can detach and reattach components.
Be aware that all incubation components will continue working with the
Incubator
tool window
specified parameters after ZEN has been shut down or has stopped working. This will keep your
sample alive, even if ZEN stops unexpectedly during an experiment. To turn off the incubation,
uncheck the corresponding checkboxes in the
Incubator
tool window or turn off the Incubation on
the remote control.
You will have access dependent on the attached equipment to
−
Temperature (°C) control
−
Atmosphere (%) control
