To edit any of the input boxes, press the
Override
button. To get back to the settings provided by the
software, press the
Master Reset
button.
In the
Microscope Parameters
section are the most important system parameters for PSF generation
that are not channel-specific displayed.
You can select the system used for data acquisition in the
Microscope dropdown
list. There are three
options: Single plane illumination microscope, conventional microscope (also known as a widefield
microscope), and confocal microscope for which the additional pinhole diameter parameter applies.
Dimension:
You can view and change the numerical aperture for the detection, which results as a combination of the
detection optic and the used zoom setting, in the
NA Obj.
input field. So, it is not the numerical aperture
of the detection optic alone which is indicated on the lens itself or displayed in the software.
The
Immersion input
field displays the refractive index of the immersion medium. Please note that this
can never be smaller than the numerical aperture of the objective. You can make a selection from typical
immersion media in the dropdown list next to the input field.
The
Scale lateral
input field displays the geometric scaling in the X/Y direction.
The
Scale axial
input field displays the geometric scaling in the Z direction.
When scaling in this window is changed it will override the settings chosen in the
Multiview Processing
window (Fig. 233).
In the
Channel specific
section, you will find all settings that are channel-specific. This means that they
may be configured differently for each channel.
The
Illumination
input box holds the excitation wavelength for the channel [in nm]. The maximum is
assumed. The color field corresponds to the wavelength (as far as possible).
The
Detection
input box represents the maximum of the emission wavelength for this channel dye. The
color field corresponds to the wavelength (as far as possible).
The
NA Lightsh.
Input box displays the numerical aperture (NA), which is resulting from a combination
of the illumination optics and the zoom settings used during image acquisition.
