8.0 Protocols
The Corning® Ascent™ FBR system provides users the flexibility to customize experiments. Users select the programmed Sequences under
Media, Harvest and/or Auxiliary Phases to achieve their experiment needs. Media Phases can run after a Harvest Preparation Phase but
not during an active Harvest Phase. Some phases can only run once while other phases can run multiple times. As an added benefit, up to
two Auxiliary Phases can be introduced during system set up. (Table 6-1 in Section 6.0: Process Overview).
This section describes steps and sequences in the Media, Harvest and Auxiliary Phases (Figure 8.1). Use Table 8-1 as a quick reference
guide to view specific phase protocols. Detailed machine sequencing for each phase can be found in Appendix 3.
NOTE:
In situ lysis is not a programmed phase. A manual protocol can be found in Section 8.5.

• Auto Resume: To be used for temporary power loss (at user’s discretion).
All equipment modules are resumed at their previous setpoints and outputs. Control modules not controlled by the equipment
modules will return to off. Phase progress is lost, and the user must initiate a phase to continue.
• Manual Resume: For extended power loss or safety condition (at user's discretion).
All control modules, equipment modules are off, and the previous phase is aborted. User must manually initiate control modules,
equipment modules, and phases to proceed.
• Shutdown: Proceed to Shutdown Phase.
All control modules and equipment modules are off, and the previous phase is cancelled. Shutdown Phase is run immediately.
There is no data loss in the case of power loss. The system retains the data that was logged prior to power loss in the data file, and
resume data logging (if shutdown is not selected) after power restoration.
