Appendix 3: Operational Sequence Reference Tables
This section provides detailed reference tables to the instrument state and operational sequences. Start with the User Action in the
left column which triggers the system sequence states described in the right column.
1. Media Phases
Initialize
User Action
System Auto Sequence
User selects Initialize at the HMI.
Prompts user to enter Batch ID.
User enters Batch ID.
Start batch timer and begin data logging. Prompts user to confirm
that pump calibration is complete, if applicable.
User calibrates pumps offline, as required.

User scans the MCV and bioreactor bar codes
and installs the MCV and bioreactor, connects
sensors, and verifies the following parameters:
DO and pH Calibration Values and the BRX Loop
Volume.
Bar code parameters automatically populate at respective setpoints.
System waits for the user to confirm set up completion.
Prompts user to confirm completion of setup and verify mass flow
controllers and pumps are turned off to zero the pressure sensor.
User completes Manual setup and confirms
prompt.
De-energizes gas shut off valve and turn pumps off.
Allows the user to select zero-point on the main HMI screen next to
pressure sensor.
Prompts user to perform zero-point adjustment of pressure sensor
and confirm when complete.
User selects zero-point adjustment at HMI and
confirms prompt.
Performs global reset on all alarms. Clears alarm history.
Removes zero-point button at the HMI.
Resets all pump and mass flow controller totalizers. Sets Current MCV
Volume to 0.
De-energizes orange and red pinch valves for 5 sec.
Energizes orange and red pinch valves to fully actuate.
Prompts user to verify that orange and red pinch valves are fully
actuated.
If the user selects Yes, proceeds to the next step.
If the user selects No, repeats valve actuation steps once.
User inspects pinch valves and selects prompt
response.
De-energize orange and red pinch valves.
Turn exhaust heater on.
Initialize phase button is grayed out.
Prompt operator that system setup is complete.
User acknowledges initialize is complete.
Media Fill and Prime
User Action
System Auto Sequence
User bar code scans Media Addition bottle and
verifies data.
Bar code parameters automatically populated to applicable control
module and phase setpoints.
User initiates Media Fill/Prime at the HMI.
User connects fresh media bottle (e.g., 3L
Erlenmeyer flask via AseptiQuik® connectors to
the media transfer line for filling the MCV.
Phase window pops up for the user to enter the following setpoints:
1. Media Flow Setpoint (mL/min.)
2. Media Fill Volume (mL)
3. Recirculation Pump Flow Setpoint (mL/min.)
The recirculation consumable volume (mL) is auto populated after bar
code scanning.
User enters setpoints for a 2.5 m

bioreactor.
For example:
• Media Flow Setpoint 300 (mL/min.)
• Media Fill Volume 1300 (mL)
• Recirculation Pump Flow Setpoint
50 (mL/min.)
Recirculation Consumable Volume 550 mL is
automatically populated.
User selects the play button.
Orange pinch valve is de-energized.
Red pinch valve is energized to open to the media container.
Media pump starts clockwise at the MCV Fill Pump Flow Rate setpoint
and stops when the totalizer reaches the MCV Fill Volume.
Adds Media Volume from totalizer to the Current MCV Volume.
Prompts user to confirm that the displayed Current MCV Volume on
the HMI is correct.
User visually checks media level in the MCV
and confirms Current MCV Volume.
Prompts user to confirm Media Fill is complete or select Media
Addition Phase.
