Experiment Run
An experiment run takes place after Initialize phase is completed.

Select Media Fill/Prime phase.
Section 8.2.2
• Connect the media tubing lines in the orange and red valves.
• Connect the Media Addition bottle via AseptiQuik® G connectors. Connectors must audibly click when
mated. Pull out the (blue) tabs.
• Manually clamp the unused AseptiQuik® tubing lines.
• Fill the MCV.
• Enter setpoints. Volume will vary depending on bioreactor size.
• Check liquid level in the MCV using gradations on the vessel. Confirm or adjust current MCV volume.
• Start Media Prime.
• De-air the bioreactor. Jog pump in Operator mode for approximately 1 min. at 500 to 700 mL/min. to
purge remaining air bubbles, both clockwise and counterclockwise direction.

Select Media Conditioning phase to stabilize temperature, DO and pH.
Section 8.2.3
• Verify the system is stable (2 to 4 hours minimum) or preferably overnight. Verify system is stable in
Trends.
• Perform Sensor Calibration.
• DO sensor calibration.
- Enter a new reference value (typically 100% of air saturation).
• pH sensor calibration.
• Take a media sample and measure pH level offline. Enter the new pH Reference Value.
• Temperature sensor calibration
- MCV temperature offset is typically 3°C.
- FBR temperature offset is typically 0.5°C.

Select Media Maintenance phase. Enter parameters:

• MCV TIC (Temp) setpoint 0 to 70°C, typically 35°C to 42°C
• FBR TIC (Temp) setpoint 0 to 70°C, typically 35°C to 42°C
• MCV DO setpoint 0 to 200%, typically 100%
NOTE:
The system does not allow for setpoints below 100%.
• FBR DO setpoint 0 to 200%, typically 15% to 30%
• Min. CO2 % 0 to 30%, typically 4% to 6%
• pH setpoint 6.0 to 8.0, typically 7.0 to 7.2 pH

Select Inoculation phase.
• Connect inoculum bottle to the Feed line.

• Add Inoculum Volume to Current MCV Volume and to Current Total System Volume.
• Sample media to monitor attachment kinetics (every 15 minutes for the first hour; every 30 minutes
for the subsequent 2 hours).
• Use the system volume to calculate the total number of cells attached.

Add Base.
Section 8.2.5.3
• Install Base tubing in base pump (yellow/blue clips).
• Prime Base tubing. Stop pump (in manual mode) before base enters MCV (approximately 15 to 20 mL).
Return the pump into Program mode.
