NOTE 1:
The bioreactor is re-primed at the Recirc Pump Addition Flow Rate. The Volume Sent to
Bioreactor is automatically set to the BRX Loop Volume regardless of whether media was removed
from the bioreactor.
NOTE 2:
The system automatically limits the Media Pump Addition Volume to prevent sending a
volume to the MCV which causes it to exceed the fill limit. The Media Pump Addition Volume field
flashes red and white as a warning.

User confirms the Current MCV Volume at the HMI.

A prompt to complete manual actions will display. User shall manually perform the following:
- Clamp used media addition line.
- Install a new media addition bottle via AseptiQuik® G connections on the media addition line.
- Install the media waste bottle via AseptiQuik® connections on the media waste line.
Press
when all manual operations are complete.

A prompt will remind the user to perform tasks to drain media from the FBR.
- Exit the task prompt (press X).
- Open the valve on the return media line.
- Manually jog pump in reverse to empty the bioreactor to a desired level.
• Remember to close the valve once the bioreactor has reached the desired level.
• Automatic media removal is managed to avoid overfilling the MCV from the BRX loop by
considering the media levels and phase setpoints.
• Automatic media addition and BRX loop priming is managed to prevent air from being
introduced to the FBR during the prime.
• After the BRX Loop is re-primed, the Recirc pump ramps to the pre-Media Exchange flow rate
over 120 seconds. Once this ramp and the media addition are complete, the phase resumes.
6.
When media addition and BRX prime is complete, a prompt will ask the user to confirm the
displayed MCV volume is correct.
If the volume is not correct, press the MCV Vol Adjustment button and enter the correct volume.
