8.4.1 Harvest Preparation
Figure 8-7 shows the Harvest Kit set up in front of the Corning Ascent FBR system. The kit is made up of 5 bottles with tubing lines that
shall be installed in the respective pinch valves.
NOTE:
Pinch valves will not actuate properly if proper-sized tubing is not installed in both locations of each valve.
TIP 1:
Each bottle tubing line has a designated color clip that corresponds to the same colored pinch valve for install. Follow the color
designated on the pinch valve cover for correct tubing placement.
TIP 2:
Load tubing with the white clip into bottom valve position (next to the instrument), then load tubing with the black clip in top valve
position (farthest away from the instrument).

1. Scan harvest kit consumables.
Scan bar codes on the Harvest Kit sterile packaging, and remove the consumables from the bag.

Fill the Harvest Wash bottle with DPBS and Harvest Solution bottle with Accutase® or other proteolytic reagent in aseptic conditions.

While the harvest consumables kit is in the sterile cell culture hood, make sure the flush bottle
is tightly closed before installing the bottle into the Harvest Kit.
• Unscrew the flush bottle cap and check that both the long and short tubing on the underside
are not kinked.
• Verify that the O-ring fits well.
NOTE 1:
The long tubing should be attached to the tubing containing the blue/white clips.
NOTE 2:
The short tubing should be attached to the tubing containing the green/black clips.
