3. Drain DPBS wash from the bioreactor to the MCV.
• Open the air valve on the media return line.
• Set the recirculation pump at 500 mL/min. (reverse flow) to drain the bioreactor, Lynx® connector, and tubing into the MCV.

Sampling option.
Pull a sample from the sample collection port during the recirculation.
5. Collection.
• To separate lysis buffer from PBS wash, connect a new bottle to the Media Waste AseptiQuik® connection. An adaptor may need to be
attached earlier in the run to have appropriate connections.
• Go to Media Removal Phase, press
to pump DPBS wash from the MCV to a Media Removal bottle
8.5.4 High Salt Wash (Optional)
Add High Salt (150 mM NaCl) Wash to the MCV in the volume listed below for the following setup:
• Lynx® connector to bioreactor: 1 m

(650 mL); 2.5 m

(800 mL); 5 m
2
(950 mL)
• AseptiQuik® G connector to bioreactor: 1 m
2
(500 mL); 2.5 m
2
(600 mL); 5 m

(700 mL)
1. Ensure proper connections.
• There are several options to add lysis buffer to the MCV.
• Connect to the luer fitting dip-tube to the luer port on the MCV head plate. Apply positive pressure to the dip-tube to allow the solution
to flow into the MCV.
• Use AseptiQuik® connectors or tube weld on to the feed addition line of the consumable to connect the lysis buffer container.
TIP:
Ensure the flexible tubing is in the pump head, and not the silicone portion. An adaptor may need to be attached earlier in the run to
have appropriate connections.
• Use an unused media addition line and connect via AseptiQuik® connector.
2. Perfuse salt solution through the bioreactor.
• On the HMI, go to Bolus Addition, press to add High Salt Wash.
• Or on the HMI, go to Media Addition, and set parameters to add High Salt Wash to the MCV.
• In Operator mode, set the recirculation pump at 500 mL/min. counterclockwise (forward flow) for 30 min.
3. Drain High Salt Wash from the bioreactor to the MCV.
In Operator mode, set the recirculation pump at 500 mL/min. (reverse flow) to drain the bioreactor, Lynx® connector, and tubing into the MCV.

Sampling option.
Pull a sample from the sample collection port during the recirculation.
5. Collection.
• Connect the Collection bottle to the Media Waste AseptiQuik® port.
• Go to Media Removal Phase, press
to pump the salt wash from the MCV to a media waste bottle.
8.6 Shutdown Procedure
This sequence phase is used to safely shut down the system once the experiment is complete. Once the batch has ended, the system
will generate the process file where information from the experiment are stored. After the Shutdown Phase is completed, all phases may
run again.

Shutdown may be selected on Media Phases or Harvest Phases. Select Shutdown.
In this example, after harvest collection, the user may perform a system shutdown.

Press
to run Shutdown Phase.
3. The steps below are performed automatically by the system:
• Stops automated and manual control of all equipment modules and control modules.
• Closes batch process file.
• Ends Batch Timer.
