Tips and Applications

Using the Washer for ELISA Based Applications

Objective: wash effectively to get a dry plate to achieve low background 
and improved signal. 
All ELISA assays differ in their binding properties. Biochemical antigen-
antibody interactions can be strong or weak. 
In general, use: 

•

Medium to high dispense rate 

•

Medium to high aspiration rate 

•

Fast aspiration descent speed 

•

Long aspiration time 

•

Probe height set to 0 mm to 0.2 mm or bottom of the well 

•

Aspiration offset to the center of the well, or crosswise 
aspiration for a completely dry well 

•

Slow to fast plate shaking 

•

Bottom or Top washing 

Using the Washer for Cell Based Applications

Objective: wash effectively without lifting cells off the well bottom to 
achieve a low background and improved signal.
All cells differ in their adherent properties. For example, CHO cells 
adhere strongly, while HEK293 cells tend to adhere weakly. In general, do 
not wash your cells any more than is necessary to achieve low 
background and sufficient signal.
Different cell lines will tolerate different wash conditions. The following 
are general recommendations, but each cell line and wash solution will 
require optimization by the user.
