Pretreatment (Hemolysate Preparation)
1. Using a calibrated positive displacement micropipette,
dispense 400 µL Hemoglobin Denaturant into a
microcentrifuge tube.
NOTE:
Dispense Hemoglobin Denaturant directly
to the bottom of the tube; do not allow the
denaturant to drip down the inside of the tube.
2. Using another calibrated positive displacement
micropipette, withdraw exactly 10 µL of the well-mixed
whole blood patient specimen or control.
3. Insert the pipette into the microcentrifuge tube
containing the Hemoglobin Denaturant, allowing the tip
of the pipette to barely make contact with the surface of
the denaturant. Dispense the specimen (1:41 dilution).
4. Withdraw and redispense the mixture two times, keeping
the tip of the pipette in constant contact with fluid in the
tube.
5. Mix by gently vortexing at medium speed 5 to 10 seconds.
Avoid foaming.
6. Prior to testing, allow the hemolysate to incubate for at
least 5 minutes at 15 to 30°C.
7. Transfer hemolysate to a sample cup and place on the
instrument.
